Nelfinavir inhibits KSHV virus production. The effect of nelfinavir on KSHV virus production was examined using a standard dose-response assay. 5r219 cells were first incubated with media containing 1 µg/ml doxycycline for 1 h. After that replicate cultures were incubated with varying concentrations of nelfinavir in the continued presence of doxycycline for 72 h. Uninduced (Unind) cultures were also examined in parallel. The culture supernatants were harvested, clarified and used in qPCR assays to enumerate viral genomes (A) as well as concentrated and the pelleted virus used to infect Vero cell monolayers (B).
